Who are the Big Six?
While checking on that expression, I found a web site that explains it nicely:
Who are the Big Six, on fictionmatters.com
They are basically the six largest publishing houses in the business. Those people want to get books published with, because they make big sales. Fictionmatters.com explains them and their history. You'll find many familiar names of imprints there.

The thing to remember is that these aren't publishing houses so much as publishing groups. Each of them owns so many imprints that the vast majority of published authors are in some way associated with one of those names, even if it isn't immediately apparent by looking at the covers. I look behind me at my bookcases now and I see lots of different company names all over the shelves, each (well, most) of which still exist as distinctive publishing houses (each with their own specific 'remits'; some would be more fiction-friendly than others), but a lot of them would still be counted as being part of 'the big six'. And despite being owned by the same 'big one', these distinctive houses are still in open competition with each other (which can lead to hilarious situations when, for example, one part of Random House sues another for copyright! ).
So yes, those names are certainly ones that we've all heard, but it would be dangerous for an author to limit their aim solely on those names; if you submit to 'Random House' you'd probably still end up being published under a 'different' publisher (though it should be noted that the author's agent should be the one doing the submitting, and in theory they'd know who they'd be sending stuff to and why).
